dc.description.abstractThe aim of this study was to compare plasma rico en plaquetas (PRP) obtained from blood of cows using sodium citrate (SC) or citrate, citric acid, dextrose-solution A (ACD-A) anticoagulants regarding the content of platelets and leukocytes (WBC), and the degree of platelet activation in whole blood (WB) and (PRP). Blood samples were collected from 15 clinically healthy Holstein cows in tubes with SC and ACD-A. Samples were processed by a double centrifugation tube method for producing PRP. PRP and WB were analysed for hemogram and automated determination of mean platelet volume (MPV) and platelet distribution width volume (PDW). Platelet counts differed (P < 0.0001) between WB and PRP for each separate anticoagulant. The concentration of platelets in PRP was 2,3 and 2,5 times for SC and ACD-A, respectively, compared to platelet counts in WB for both anticoagulants. PRP platelet concentration did not differ between both anticoagulants. Values for MPV and PDW were similar in PRP obtained after centrifugations regardless of the anticoagulant used. Leukocyte counts were higher (P < 0.001), particularly lymphocytes, in PRP obtained with SC in comparison with ACD-A. In conclusion, results obtained allowed to establish that there are no differences for obtaining bovine PRP, using either SC or ACD-A as anticoagulants. The results suggest platelet conservation homogeneous and structurally stable in bovine PRP, especially when sodium citrate was used.
