dc.description.abstractEl uso abusivo de fármacos ha provocado la aparición de resistencias antihelmínticas en ésta y otras especies de nematodos, motivo por el cual se han considerado diferentes estrategias de inmunoprotección como herramientas alternativas para el control de las infecciones gastrointestinales por este nematodo. En este sentido, son numerosos los estudios que han demostrado la capacidad protectora de enzimas proteolíticas de vermes adultos de H. contortus. Con el objeto de analizar las interacciones entre edad e inmunoprotección, en el presente trabajo se realizaron ensayos guiados de inmunización en ganado ovino canario de 3 y 6 meses con este tipo de antígeno. Se inmunizaron cinco ovinos de 3 y 6 meses de edad con dosis crecientes de inmunógeno durante las 5 primeras semanas de la experiencia, considerándose sus correspondientes grupos control. A la 7ª semana del inicio del estudio los animales se inocularon con 7.000 L3 de H. contortus vía intra-ruminal. En los vermes de los diferentes grupos de animales recogidos tras el sacrificio se llevó a cabo un estudio de variabilidad genética mediante Single Strand Conformation Polymorphism (SSCP) en genes codificadores de proteinasas tipo cisteína (genes AC-1, AC-3, AC-4 y GCP- 7) con el fin de valorar posibles variaciones en las frecuencias génicas de los vermes recogidos de animales inmunizados de 3 y 6 meses. Los resultados pusieron de manifiesto la efectividad de la inmunización en los ovinos de 6 meses, con una reducción de los recuentos de huevos en heces y del número de vermes en abomaso. Así mismo, a nivel genético, y en concordancia con el nivel de protección descrito, se observaron diferencias estadísticamente significativas en todos los genes analizados al comparar los vermes recogidos de animales inmunizados y no inmunizados de 6 meses; los alelos cuya frecuencia alélica se vio disminuida tras la inmunización podrían considerarse como alelos codificadores de proteinasas que estarían involucradas en el proceso de inmunoprotección. Una variabilidad genética similar pudo observarse en algunos genes en los vermes procedentes de animales de 3 meses, a pesar de que no pudo demostrarse una clara protección en los animales inmunizados a esta edaden_US
dc.description.abstractThe extensive use of drugs has contributed to the emergence of anthelmintic resistance against this and other species of nematodes, so different strategies to control gastrointestinal helminth infections have been considered as alternative tools. In terms of immunoprophylaxis a number of studies have demonstrated the protective effect of proteolytic enzymes of adult worms of Haemonchus contortus. In order to analyze the interactions between age and immuronoprotection, different immunization trials with this type of antigens using Canarian sheep 3 and 6 month old were performed. Five sheep were immunized at 3 and 6 months of age with increasing doses of antigen for the first 5 weeks of the experiment, while buffer was given to the corresponding control groups. At week 7 of the beginning of the study, animals were challenged with 7,000 L3 of H. contortus via intra-ruminal. In worms from the different experimental groups a polymorphism study of genetic variability was carried out by Single Strand Conformation Polymorphism (SSCP) in genes encoding cysteine proteinases (genes AC-1, AC-3, AC-4 and GCP-7) in order to investigate alelic variability in worms harvested from immunized animals aged 3 and 6 months. The results showed the effectiveness of immunization in 6 month old sheep, with a reduction in faecal egg counts and the number of worms in the abomasum. Also, at the genetic level, in accordance with the level of protection, statistically significant differences were observed in all the genes analyzed by comparing immunized and non-immunized animals of 6 months of age; the alleles whose frequencies was decreased after immunization might be considered as alleles encoding proteinases that would be involved in the process of immunoprotection. A similar genetic variability was observed in some genes in 3 month old animals, despite a clear protection in immunized animals at this age could not be demonstrateden_US
