Abstract: Protoplasts isolation is a potential way to experiment with the gene expression in algae as has been done previously in higher plants. Protoplast isolation from Bonnemansonia hamifera and Antithamnion decipiens was developed following the optimization of protoplast isolation protocol for obtaining free cells in the better conditions possible. Furthermore these naked or semi naked cells will be tested as gene expression system in order to study the functionality gene. The enzyme type and concentration, the incubation period, temperature and tissue source on the protoplast yields were used to optimize the isolation procedure The enzyme cocktail which gave the better protoplast structures was a mixture with 3% cellulase and 6% hemicellulase. The free cells were cultured in ASW and PES medium.
